PhD Position in Bioproduction of Genetically Modified Monocytes
Develop robust, GMP-compliant bioprocessing methods for the production of therapeutic monocytes to treat triple-negative breast cancer.
Best for:Best for candidates with a background in bioprocess engineering, cell therapy, or biotechnology interested in GMP-compliant manufacturing.
Overview
This PhD project is part of the 'Mono-Chem' PEPR Biotherapies Bioproductions initiative, focusing on the development of genetically modified human monocytes as therapeutic vectors for triple-negative breast cancer. The successful candidate will work within the MTInov integrator, a collaboration between the CHRU of Nancy and the Laboratory of Reactions and Chemical Engineering (LRGP). The research involves optimizing cell isolation and transduction processes under Good Manufacturing Practice (GMP) conditions using automated systems like Sepax C-Pro and CliniMACS Prodigy. The goal is to establish scalable, robust protocols that ensure high cell purity, viability, and therapeutic efficacy for clinical transfer.
What you'll do
- Develop and optimize scientific methodologies for the isolation and transduction of monocytes under GMP conditions.
- Compare and evaluate different automated isolation methods using the CliniMACS Prodigy system.
- Optimize transduction protocols using lentiviral vectors in bioreactors such as G-Rex or CentriCult.
- Ensure all developed processes meet strict quality specifications for purity, viability, and modification rates.
- Perform functional qualification tests on modified monocytes and macrophages.
- Collaborate with multidisciplinary teams to facilitate the clinical transfer of therapeutic cell products.
