PhD Thesis in Large-Scale Proteomics and Structural Mass Spectrometry
Develop next-generation inhibitors targeting proteasome subtypes for autoimmune and cancer therapies.
Best for:Best for molecular biologists, structural biochemists, or proteomics specialists with experience in mass spectrometry and inhibitor design.
Overview
This PhD position at the Institut de Pharmacologie et Biologie Structurale (IPBS) focuses on advancing third-generation proteasome inhibitors to selectively target proteasome subtypes associated with PA28 activators. The project aims to reduce toxicity linked to current treatments for autoimmune diseases and cancer by developing highly specific inhibitors for complexes like i20S-PA28αβ and c20S-PA28γ. The work combines structural mass spectrometry, proteomics, and cell-based assays to evaluate inhibitor efficacy, specificity, and toxicity. The successful candidate will contribute to groundbreaking research with potential clinical applications in autoimmune diseases, inflammatory conditions, and cancer treatment.
What you'll do
- Develop and evaluate third-generation proteasome inhibitors targeting i20S-PA28αβ and c20S-PA28γ complexes.
- Conduct in vitro and in cellulo assays to assess inhibitor efficacy, specificity, and toxicity.
- Perform large-scale proteomics and structural mass spectrometry to analyze off-target effects and binding specificity.
- Identify and characterize peptides that reflect inhibitor binding to target and off-target proteins.
- Study the mechanism of action of inhibitors, including effects on proteasome disassembly and stoichiometry.
- Publish findings in high-impact scientific journals and present results at international conferences.
- Collaborate with interdisciplinary teams to integrate findings into broader drug discovery pipelines.
